Antonios G. Mikos

Louis Calder Professor of Bioengineering and Chemical Engineering; Professor of Chemistry

Our laboratory emphasizes the use of synthetic biodegradable polymers as supportive scaffolds for cells, as conduits for guided tissue growth, as specific substrates for targeted cell adhesion, or as stimulants for a desired cellular response.

We are investigating bone regeneration and repair using a biodegradable polymer scaffold either by inducing bone tissue growth post-implantation or by seeding the scaffold with bone cells prior to implantation. We are developing new processing methods for manufacturing three-dimensional polymer scaffolds including combinations of solvent-casting or extrusion-molding with particulate-leaching techniques. We are further fabricating injectable, in situ polymerizable, biodegradable composite scaffolds for bone replacement also serving as carriers for bone cells and growth factors to induce a bone regeneration cascade.

We are examining the effects of mechanical forces and flow on three-dimensional cultures of bone cells on the gene expression and the production of molecular mediators of mineralization and bone formation including extracellular matrix components and growth factors. We are also developing new CAD/CAM and polymer chemistry fabrication technologies for the production of synthetic biomimetic materials that exhibit the mechanical responsiveness and biochemical processing capabilities of living cells and tissues.

We are synthesizing new injectable, in situ crosslinkable, biodegradable poly(ethylene glycol) copolyesters with covalently bound cell adhesion molecules for modulating cellular function to the hydrogel. We are designing new proactive hydrogels conducive to bone ingrowth but preclusive to fibrous infiltration for treating dental defects with guided tissue regeneration. Moreover, we are proposing the use of injectable biodegradable hydrogels after interventional cardiovascular procedures to repave and heal injured arteries and prevent arterial restenosis. We are fabricating thin films of biodegradable polymers with patterned surface chemistries and morphologies in an attempt to modulate the function of cultured cells with the ultimate goal of regenerating retinal pigment epithelium. We are manufacturing porous biodegradable polymer conduits for peripheral nerve regeneration to serve not only as guidance scaffolds but also as carriers for nerve growth factors and cells. Finally, we are developing non-viral gene delivery systems using synthetic polymers with targeting moieties for gene therapy.
